What is Addyi?

Addyi®, also known as Flibanserin, is a multifunctional serotonin agonist and antagonist (MSAA) that improves sexual arousal and functioning in premenopausal women who are suffering from reduced sexual desire and interest. This non-hormonal pill is taken once daily at bedtime.

Conditions treated by Addyi

Addyi is used to treat premenopausal women with hypoactive sexual desire disorder (HSDD), which is characterized by low sexual desire that causes marked levels of distress or interpersonal difficulty, and is not due to a co-existing psychiatric or medical condition, relationship problems, or the effects of medication or other drugs.

This medication is not prescribed to men and it is not used for the treatment of HSDD in postmenopausal women. Furthermore, this medication is not used for the sole purpose of enhancing sexual performance.

What are the side effects of Addyi?

The most common side effects that are associated with taking this medication include:

  • Dizziness
  • Somnolence
  • Nausea
  • Fatigue
  • Insomnia
  • Dry mouth

Less common side effects include the following:

  • Rash
  • Anxiety
  • Vertigo
  • Sedation
  • Constipation
  • Abdominal pain
  • Metrorrhagia (Abnormal uterine bleeding)

Appendicitis is a rare but serious complication that may occur during treatment. In addition, sleepiness may arise, especially during waking hours for women who drink alcohol, or take certain medication and/or herbal supplements. Hypotension (Low blood pressure) and fainting may occur if this drug is taken during the day, if alcohol is consumed within two hours of taking it, or if it is taken with herbal supplements or other medicine.

Avoid using grapefruit juice or St. John's wort with Addyi. Tell your doctor before starting any new prescription or over-the-counter medication while using Addyi.

If feeling faint or lightheaded, then lie down and call for medical help.

What you should know before using Addyi

The following factors should be considered before taking Addyi (flibanserin): The combination of alcohol and this drug increases the risk of low blood pressure and fainting. Patients should be counseled about the importance of separating alcohol consumption and the use of Addyi by at least two hours. Wait at least two hours after consuming one or two standard alcoholic drinks before taking Addyi at night. After this drug is taken, alcohol should not be consumed until the next day. Do not use Addyi if three or more alcoholic beverages were consumed within 6 hours prior to bedtime.

Severe low blood pressure and fainting can occur when this medication is taken along with moderate or strong CYP3A4 inhibitors, Strong CYP2C19 Inhibitors, or oral contraceptives, as well as in patients with hepatic (liver) impairment. As a result, it is not recommended in such cases. Taking Addyi increases digoxin levels and this may lead to digoxin toxicity. Therefore, digoxin levels should be closely monitored in order to detect increased concentrations.

The risk of experiencing low blood pressure, somnolence, sedation and fainting may occur from taking Addyi alone and medical attention is promptly required if the symptoms do not resolve or if they worsen.

After taking this drug, patients should avoid activities that require full alertness (e.g., driving or operating machinery) for at least six hours until the manner in which this drug affects the individual is fully recognized.

Individuals who are taking the following medications should not use this drug:

  • CYP3A4 Inducers
  • Nefazodone, which is a medication that is used to treat depression
  • Certain medications that are used to treat HIV-1 infections
  • Certain oral medications that are used to treat fungal infections
  • Certain antibiotics including: ciprofloxacin, telithromycin, erythromycin, clarithromycin
  • Certain medications that are used to treat Hepatitis C infections
  • Certain medications that are used to treat chest pain (angina), high blood pressure, or other heart problems

This medication is not recommended for nursing mothers. There are no studies of ADDYI in pregnant women to identify the potential risk to unborn babies. However, animal models indicate that the risk for potential harm to an unborn baby cannot be ruled out.

If a dose of this drug is missed at bedtime, the next dose should be taken at bedtime on the following day. A double dose should not be taken.

If alcohol is consumed in the evening, skip the dose at bedtime and take the next dose the following day at bedtime. Alcohol should not be consumed after this drug is taken.
