What is Lynparza?

Lynparza (also known by its generic name Olaparib) is a medication that is used for treating certain types of ovarian and breast cancer: BRCA-mutated ovarian cancer, and BRCA-mutated HER2-negative metastatic breast cancer.

Many types of cancers are caused by damaged or mutated genes in our DNA. Therefore, it makes sense to search for therapies that address the damaged or mutated DNA that cause cancer. Lynparza is the first of a new class of medications – known as PARP inhibitors – that does exactly that.

About 15% of ovarian cancers and 5-10% of breast cancers are caused by mutations in specific segments of DNA, known as BRCA genes. The BRCA gene mutations cause DNA defects in the cancer cells, which must be repaired for the cancer cells to proliferate and survive; Lynparza inhibits these cancer cells' ability to do that.

Lynparza currently is used for treating specific types of ovarian and breast cancer, as well as some peritoneal, fallopian tube, and pancreatic cancers.

What are the side effects of Lynparza?

The most concerning Lynparza side effects observed in clinical trials were:

  • Serious blood disorders (myelodysplastic syndrome and acute myeloid leukemia) occurred in less than 1.5% of people taking Lynparza, with most cases being fatal
  • Pneumonitis (inflammation of the lungs) occurred in less than 1%, with some cases being fatal

Other common Lynparza side effects include:

  • Fatigue
  • Low red blood cell counts (anemia), white blood cell counts (neutropenia/leukopenia), and platelet counts (thrombocytopenia)
  • Nausea, vomiting
  • Reduced appetite
  • Abdominal pain
  • Diarrhea
  • Constipation
  • Inflammation inside the mouth
  • Indigestion
  • Upper respiratory tract infection
  • Urinary tract infection
  • Shortness of breath
  • Dizziness
  • Distorted sense of taste

What you should know before using Lynparza

Lynparza can cause fetal harm, so the drug should not be taken by women who are or may be pregnant. Women of reproductive potential should use effective contraception during treatment and for six months following the last dose. Women should not breastfeed while taking Lynparza, and for one month after the last dose.

People should not eat grapefruit or Seville oranges, or drink grapefruit juice or Seville orange juice while taking Lynparza.
